Introduction: GS-441 is a competitive inhibitor of nucleoside triphosphate. It can inhibit the replication of several different types of RNA, such as severe acute respiratory syndrome (SARS), Middle East respiratory syndrome, Ebola, Lassa fever, Junin and respiratory syncytium, etc., and has low cytotoxicity in a wide range of cell lines. Function: Gs-441 is phosphorylated to nucleoside monophosphate (NMP) by cell kinase, and then transformed into an active triphosphate metabolite (NTP structural analogue). In RNA synthesis, active NTP structure analogues act as natural nucleoside triphosphate competitors and compete with natural nucleosides (ATP, TTP, CTP, GTP) to participate in RNA transcription. When gs-441 is inserted into the transcripts, transcription will be terminated early, thus inhibiting the RNA transcription process.
